The black line is probably just a random, non-recurring ink streak. The USA 29 was printed separately from the other colors, and slight shifting around, relative to the other colors, from printing to printing is common. Bi-color stamps with vignettes such as the 1901 Pan-American Exposition Issue or the 1918 3-cent airmail issue (#C3) with mis-registered vignettes are of high interest.
