Does having the postmaster's signature on a cover add desirability or value?

I would not think so, unless the PM was famous for something. I have a HPO cover from Yakima, Wa., that was signed by the PM of the time, Tom Stave. I bought it because when I first went to work for the Yakima PO, he was the postmaster there. That was long ago.

I can't think of any situation where it would add to the value.

I'd take one signed by Benjamin Franklin. Ya, I know he was Postmaster General, not technically a working postmaster...but still it'd be nice!

I think it depends a lot on the time period. 19th century free franks by PM's have some modest value. In the 30's, a lot of PM's signed first flights and airport dedication covers, those don't seem to make much difference. Farley signed everything, but he is famous enough that it might bring a small increase in value.

I would definitely pay more for a cover with his signature, especially if it had some of the Follies on them.

Revcollector, these are the ones I find in my collection.
