It's definitely not a serial number for the envelope. These were produced in huge quantities (my guess is over 1 million). I still have several blank ones sitting around somewhere.
It's more likely stamped/printed on the cover to indicate the quantity that was in the box (i.e., 500 cacheted envelopes in the box).
