No, I do not think it is a guide dot. Looks like some sort of blemish on the plate to me. I onder if more of these have been found

This looks most likely to be a marking on the plate with this stamp being from the rightmost column of the sheet. The other possibility is that a foreign inked object touched the edge of the sheet.

Jimjung has a point...Defective paper pulled off a full sheet temperately attached itself yo th printing process and then fell off.
