This is MiNr 5Ia, 1867 (grober Bdr,i.e., coarse impression).

Can anyone decipher the writing that's printed thereon?

Looks like a handstamp in Italian: Lettere, col, dal.

vayolene beat me to it.
Not an overprint but a postmark. Mentioned in Corinphila's 2020 auction of Werner Schindler's Collection of "AUSTRIAN MAIL IN THE LEVANT" as a shipmail handstamp applied in Trieste to incoming mail from the Levant.
