Is there a program or software, that isolates a clear image of a cancel from a stamp or cover?

I've not heard of one. Are you using any image editing software at the moment?

You need to split the color channels of the image to isolate a cancel; most image applications can do this (ACDSee does not). But there is a free online resource, it is call RetroReveal http://retroreveal.org . I put together step-by-step instructions on how to use it here

mostly I just use Photoshop and its options to reduce some specific color and change contrast and so on.
Don's link is also very useful.
but if you want to use a program on your computer you can try this:
http://www.lcs-chemie.de/poststfa.htm (German product)

under "Bildanalyse" in the program you can choose a file (better not to big) and give the program the information on the cancellation color. Also other tools are helpful like the color filters.
