Let me try this question. Say you correspond with some one frequently at the end of this route and its going to have this new direct air route. and your not a collector just sending a letter. Can you walk into the post office that is having the first flight that day and send a letter on that route and would it automatically get the cachet? Would they tell you that it is getting a cachet? I'm assuming that the reason this new direct route is created is because there is enough daily business to warrant it.
That question has been previously answered, more or less. The scenario you pose wouldn't happen today, since first flight covers are no longer a thing. But "back in the day," you would ordinarily have to request the cachet. It would not automatically be applied to all mail going out on the flight in question.
