There are no records, that I know of, of the handstamp you show appearing on a US stamp of the 1869 period.
The Hiogo, Japan cancellation, which is seen on some of the stamps of the 1869 issue, doesn't have a line across it.
It looks like this: http://www.siegelauctions.com/2012/1037/1665.jpg
The cancellation on your stamp adds no additional monetary value to the stamp. On the other hand, as it is not clear, it detracts, more than adds, to the value.

PS = It is a very pretty stamp. Just this fascination with a smudge is probably not worth the effort.

PPS - The Siegel database does not have an example of the 3c with the Hiogo cancellation on it listed in any of the past sales. They have auctions many of the great 1869 collections, so one would think that if there was such known, an example would have been in at least one of these collections.

Thanks again. All these facts you have mentioned can give us a pretty clear conclusion that this 99,9% is NOT a Japan handstamp cancellation.

I agree with Chip that the cancel doesn't add value, but it seems unusual. It doesn't look like any US fancy cancels I've seen. If it was used as additional postage to another country, the cancel is not usually centered one one stamp. From what I've seen anyway. I have a lot to learn so I enjoy looking into something like this.

No answer for this one though... possibly foreign cancel.

Since I posted early in this discussion the title has been changed to JAPANESE cancel.....
I agree with chip...not a JAPAN cancel....100% sure, not 99.9%
Looks like a ordinary cancel that was smudged.
It is what it is...sorry.
A apple is a apple......a orange is a orange....

Looks many people discussing about US post office postmarks in Japan without showing it so here is the complete list:

Hiogo postmarks

Yokohama and Nagasaki

Hakodate postmark famous for being misspelled "Hakodadi"

Prahanoaki, thank you for your contribution of these scans, they don't appear in the Scott specialized and they are, of course, very useful
