I think I know the answer to this question but I'll ask anyway. Does having the block # on a single stamp increase the value?

Seems to me some US collects collect what they call - I think - pate corners - or something like that. That's just one stamp with marginal inscription. I don't see the value of it myself unless the inscription indicates which plate block it is and you need that because the stamps vary by plate number.

They are called plate number singles and are quite collectable.

Experienced stamps need a home too. I'd rather have an example that is imperfect than no example.
I collect for enjoyment, not investment.

Like other things that determine value, attached plate numbers on singles can positively impact value. Few of "things": scarcity of the plate number, census of existing examples, classic issues with numbers generally have a premium, and I'm sure there are other factors too. Personally, I like to get plate numbered singles for my collection and since I collect primarily classic issues, they generally are a bit more pricey than issues without the number.

Does a cancelled stamp with the number attached increase the value over a mint. I would think that would be a somewhat rarer occurrence.

Some rare plate numbers may only be found to exist on single stamps rather than in blocks.

Tom, a cancelled verses a mint plate number single may or may not be more valuable. Again, it depends on other factors, not just the plate number attachment. Some cancelled, scarce stamps, are worth more than their mint counter-part. That could hold true to that particular issue being more valuable with the plate number attached.
It seems there are always variables to deal with, not the least being how one preceives value. I tend to value plate numbered stamps more just because I like them.
