Most certainly a cancelled stamp. Oftentimes the clerk would use a pen to "X" out a stamp that may not have had a postmark applied. Remember, from the earliest days to the present, the post office does not have collectors in mind when doing these things; it is merely a way to ensure that the stamp is sufficiently cancelled so as to avoid its reuse.
