Here's a nice letter. The sender, William Seton, was a prominent Catholic businessman. His daughter-in-law, Elizabeth Ann Seton, will become the first U.S. saint....Here's the cover!

Very nice! In 1790, New York City was the largest City in the USA with 33,131 inhabitants. New York City was also the largest city in the USA in 2010 but a few more folks were living there that year when the population was 8,175,133.

I know almost nothing about pre-1800 US covers. Is the black straight line "New York Feb 25" marking an official post office marking or is a privately applied one? Is there a well done reference book that shows all of the known US postal markings as well as rates from that era?

It's a post office marking. The standard reference is the American Stampless Cover Catalog, which has been digitized and is available on the U.S. Philatelic Classics Society website.
