One cannot say exactly, but generally one is looking for areas which are abnormally different than the rest of an object - areas which might indicate an alteration - areas that glow bright/dimmer or not at all or a different color than the rest. It's just another tool to help understand an item.

Not sure anyone has mentioned retro-reveal yet. That can be excellent picking up differences in cancellations on and off the stamp, and other skulduggery.
