I'm a fan of Highway Post Office covers and will occasionally pull out interesting First Trips when I spot them in boxes of covers. But they all seem to be roughly the same value. Are there any routes that are actually scarce?
No doubt there are some tougher ones, but in many ways, you have answered you own question by looking at it from the value standpoint. The truly commercial HPO uses with the standard HPO cancels are much more challenging to accumulate (and not covers with collector-requested cancels).

Just guessing, but I doubt that any of these three are common.
Eureka & San Fran HPO, that didn't actually make the trip from Eureka to San Francisco in 1948 but might have made Trip 2. Not that I blame them, Hwy 101 was a nightmare north of Willits, even in 1953.
Eureka & S. F. HPO Trip 2
Mendocino's Centennial & New P. O. Dedication in 1958 got a visit from the HPO.
