I am not sure if this is postage due, or maybe a poisoner mailing and any help identifying this would be greatly appreciated.

It makes not sense to me since it's mailed from Louisville Kentucky addressed to Delaware, but has an additional Philadelphia PA postmark. Also, it looks like one stamp was removed?

It looks like it was originally mailed at Louisville, KY on Jan 5, 1863, addressed to Philadelphia Penna, where when received, someone scratched out the original address, added the forwarding address for Wilmington, Delaware, added a stamp, and put it back in the mail at the Philadelphia PO on Jan 9, 1863.

Remember, there is no free forwarding yet, so whether the letter went home and back to the Philly post office or had the original address scratched out at the PO window, a second 3 cent charge for carrying it to the new destination was required. It is more common to draw a line or two through the original address leaving it readable and add the new address to the side.
